Automated design of primers for Gibson cloning with MacVector.

- Go to FILE | NEW GIBSON/LIGASE INDEPENDENT ASSEMBLY
- Choose the strategy
- Add a vector (from your own digest or the prepared library)
- Drag a feature (e.g. a gene) from your sequence.
- Add a second (or more) fragment.
- Allow MacVector to automatically design primers, or choose your own.
- Check the coding region around junctions to confirm the correct frame is used for protein fusions. You can add spacer residues to adjust the frame.
- Click Assemble to create the final molecule.
