How do I do Gibson cloning/LIC

Automated design of primers for Gibson cloning with MacVector.

  1. Go to FILE | NEW GIBSON/LIGASE INDEPENDENT ASSEMBLY
  2. Choose the strategy
  3. Add a vector (from your own digest or the prepared library)
  4. Drag a feature (e.g. a gene) from your sequence.
  5. Add a second (or more) fragment.
  6. Allow MacVector to automatically design primers, or choose your own.
  7. Check the coding region around junctions to confirm the correct frame is used for protein fusions. You can add spacer residues to adjust the frame.
  8. Click Assemble to create the final molecule.
