The Restriction Enzyme Picker

The Restriction Enzyme Picker (RE Picker) is a floating tool for selecting and filtering Restriction Enzymes to simplify the identification of useful enzyme cut sites.

How to use the RE Picker.

  1. Open your sequence
  2. Switch to the MAP tab.
  3. In the RE Picker dialog toggle enzymes in the displayed list
  4. Change Min or Max sliders to change the number of cut sites
  5. Change End Structure to toggle between sticky ends or blunt cuts.

When the RE Picker is first opened some initial filter parameters in the RE picker are set from the global SCAN DNA FOR.. Restriction Enzymes preferences. These are the MAX CUTS and the restriction enzyme file that is used. It also initially shows only enzymes that will cut. However, you can dynamically filter what sites are displayed using many attributes.

The RE Picker window shows all of the enzymes that match the filter criteria, even if they are not currently selected for display.

Choosing a restriction enzyme file

The currently used file of restriction enzyme sites is displayed. By default this is the Common Enzymes file. However, you can click SELECT to choose a different restriction enzyme file. You can easily edit and create your own files.

Exporting cut sites

You can take a set of cut sites from one sequence as the input to digests of other sequences for easier planning of construct workflows.

Note: When the RE Picker is visible, the “mode” of the RE search always changes to “SHOW SELECTED ENZYMES ONLY”, i.e. only those enzymes visible and selected in the tab are displayed in the Map view. This may be different to the setting used by SCAN DNA FOR.. Restriction Enzymes which controls which enzymes are visible when the RE Picker is not visible.
